Besides alternate costumes for the main characters, Chris and Sheva, there are two new unlockable characters in Resident Evil 5: Jill and Wesker. Each of these characters has an alternate unlockable costume as well.
Jill Valentine
Jill Valentine was introduced in the first Resident Evil game, where she was one of the two playable characters. She was also the main character in Resident Evil 3: Nemesis and was one of the playable characters in Resident Evil: Umbrella Chronicles. Like Resident Evil 5's main characters, Jill is a part of the BSAA. Leading up to the events of Resident Evil 5 Jill was believed to be dead, but its revealed in the game that she is still alive.

Resident Evil 5 Jill Valentine
Jill Valentine, the first primary female protagonist in the Resident Evil 5 series, is the long time partner of Chris Redfield and a member of the Bioterrorism Security Assessment Alliance. Prior to this, she was a member of S.T.A.R.S. Alpha Team and during this time, she survived the infamous Mansion Incident and the Raccoon City Incident. Jill has devoted her adult life to stopping bioterrorism and Albert Wesker.
Albert Wesker
Like Jill Valentine, Albert Wesker was introduced in the first Resident Evil game as a supporting character. Early in that game, Wesker disappears but is later discovered to be a double agent working for the Umbrella Corporation. In most of the game's endings, the player witnesses Wesker's death at the hands of the Tyrant, but there is one ending where he escapes. Wesker reappeared in Resident Evil: Code Veronica and later Resident Evil 0 and Resident Evil 4.

Resident Evil 5 Albert Wesker
The primary antigonist of the Resident Evil series, Wesker is a power-hungry, intelligent and cunning enemy. He was affiliated with the Umbrella Corporation, as one of its most promising researchers, and participated in illicit activities at the same time by going undercover as an officer in the R.P.D. and the leader of S.T.A.R.S. in Raccoon City. Wesker faked his death, gained superhuman powers from an experimental virus, and worked alongside both the mysterious "Organization" and Tricell.
