It has been confirmed that there will be Big Sisters in BioShock 2. Similar to the Big Daddy enemies that protect Little Sisters in the first game, these Big Sisters have the same kind of helmet, but a better fitting diving suit, and Little Sisters on their backs that are a bit older than previous ones. Big Daddies in BioShock 2

- Big Daddies will make more than appearance in Bioshock 2, players will actually play as them. Players will take control of a renegade Big Daddy who is searching for a Little Sister of his own. When you encounter a Little Sister, you'll have the option to either harvest them or adopt them as your own. If you harvest the little girl, you'll obtain some ADAM, but if you adopt her, she'll be able to harvest ADAM from other corpses for you. If you have a Little Sister sidekick, she will also be able to warn you when a Big Sister is getting uncomfortably close.
Players will most likely get all of the tools that Big Daddies had in the first game when they were fighting you. You'll big equipped with a large hand-drill and plasmid powers, among other things. Kotaku: Spoiler Alert! How Big Daddies Appear In BioShock 2 (March 13, 2009)
