Harper Lee uses the mockingbird as a symbol of innocence, so to kill a mockingbird is to destroy innocence. Jem, Scout, Dill, Tom Robinson and Boo Radley all are identified with mockingbirds, because they are innocent people who have been injured (if not outright destroyed) by contact with wickedness and cruelty.
As Miss Maudie tells Scout: "Mockingbirds don't do one thing but sing their hearts out for us. That's why it's a sin to kill a mockingbird."
