The enterprise is a warship and the Falcon is a cargo transport. It would be like a pickup truck with a colt 45 (gun not malt liquor) in the glovebox against a tank. The Falcon is nimbler but just doesn't have the firepower to take down the enterprise.

M$Also, you have to figure that the Millenium Falcon is only armed with two radial turrets. One on the top, and one on the bottom. There are also the mini that are lined around the bottom, but those are only good for close combat against people on land, not another ship.
The Starship Enterprise has torpedoes, a tractor beam, lasers, and whatever the crew can re-engineer on a short-term basis. While it is an exploration ship, is is extremely well-armed. The Falcon would not stand a chance.
