Gotta be "Bogey and Baby" AKA Humphrey Bogart and Lauren Bacall. An enchanting couple both on and off screen.

Even though this is totally girly and cliché, I would have to say the best on screen couple is Noah and Allie from The Notebook. They are a very believable couple. They argue, fight, and don't always seem right for each other. However, in the end, they still are obviously and deeply in love. It is apparent that one would do practically anything for the other; their love defies time and unfortunate incidents.

Laurel and Hardy because they were known for their slapstick humor. It was funny to watch their arguments with each other, and although it was in black and white, it was cartoon like.

I would have to say the best on-screen couple is Brad Pitt and Angelina Jolie from Mr. and Mrs. Smith the movie. They both have great passion about one another during the making of the film and even had an off film relationship. Both of them are very hot and just work great together.

Tough call - I'd like to nominate the following couples:

A. Samantha Baker & Jake Ryan ("Sixteen Candles")
B. Baby Houseman & Johnny Castle ("Dirty Dancing")
C. Neil Page & Del Griffith ("Planes, Trains, and Automobiles")

...but since none of those starred in a SERIES of movies together, the WINNER IS.......

Well I'd say Spike and Linda from the British TV series 'Press Gang', but I'm presuming you mean a movie.

Hard to pick, but my top 3 would be:
Ruthless People, Judge Reinhold and Helen Slater
Top Gun, Tom Cruise and Kelly McGillis
Romancing the Stone, Michael Douglas and Kathleen Turner

Definitely not 'Out of Sight' with Clooney and Lopez... it was a pointless movie.

Al and Peg Bundy from Married...with Children.

Not the typical lovey-dovey perfect couple, but hilarious, and underneath all the insult-slinging, they share moments which show us that there's nobody else they'd rather be miserable with.

Oh, such a hard decision! This really comes down to personal opinion, I suppose. So, I'm going to go with the couple from one of my favorites movies, Gone with the Wind.

The couple is Scarlett O'Hara and Rhett Butler who were played by Vivien Leigh and Clark Gable. I love them as a couple because they are nothing like the quintessential fairytale couple in most romance movies. They seem real, troubles and all. Their relationship is very love/hate, but it feels incredibly genuine. Their on-screen chemistry was amazing, and I love the traditional aspect of their love story.

House and Cuddy! Yes, I know: you can easily argue that they aren't together, and therefore aren't "really" an on-screen couple. However: they want to be together so badly that the tension rolls off them in waves.

They spend most of their on-screen time torturing each other, which is a dead giveaway that they have feelings for each other - and it makes for great TV. (Then there's the fact that House just can't resist tormenting Cuddy re: her clothes and chest.)

Besides: you never know what's going to happen in the next season of "House" - they could finally be a "proper" on-screen couple.

Marcello Mastroianni and Anita Ekberg in "La Dolce Vita" of Federico Fellini.
The combination of ironic, romantic and nostalgic scenes make this couple the most beautiful in movies history.

Definetly Dana Scully and Fox Mulder from the X Files......
Although there are others such as Leonardo and Kate from Titanic I feel that Dana and Fox are for sure the best on screen couple. Without them x files would not be x files.. Simple!

Audrey Hepburn and Gregory Peck (In Roman Holiday)
Nominated for 10 Oscars and winner of 3 (including Audrey Hepburn's as Best Actress), the Academy agreed that Roman Holiday was a charming and incredibly fun film. The scenery was breathtaking and captured the beauty that Rome encompassed. With lighthearted dialogue, Audrey Hepburn and Gregory Peck played the roles with ease and comfort.
