You won't be able to discover it by yourself. As you progress in the game, you'll meet up with your uncle inside the villa. He'll show you the way into the sanctuary. It is in the working office, where the codex wall is also located.Your uncle will open up a secret wall, leading down a stairs, into the sanctuary.
