The Flintstones aired on ABC from September 30, 1960 to April 1, 1966. William Hanna and Joseph Barbara who were well known in the animation field at that time produced the series, which was set in the Stone Age.
The Flintstones was one of the first highly successful prime time animated series. As for whether this was the first time that a husband and wife were the key characters in a cartoon series I can’t find anything to dispute this claim. Yes there would be married couples in animation to follow in the tradition of the Flintstones, such as The Jetsons, which of course was also put out by Hanna Barbara.
The Flintstones remain in reruns to this day airing over on the Boomerang Network. The series spun off eleven total spin-offs including “The Pebbles and Bam Bam Show”, and “The Flintstone Kids”, but none of them were as popular as the original series.
