In general, the EOS 1DS Mark III is a professional-grade camera, made to be more durable, and with more accurate control of shots, whereas the EOS 5D Mark II is a "prosumer" camera, with many more features of interest to the amateur enthusiast. These include a higher resolution LCD screen, more movie modes, broader ISO range etc.
Some differences in specs are:
1. The EOS 5D Mark II has a broader ISO range by far.
2. The 5D-MII shoots up to 3.9 images per second vs. 5 for the 1DS-MIII.
3. The image processor is DIGIC IV for the 5D-MII vs. dual DIGIC III processors for the 1DS-MIII. The dual processors allow faster handling of rapid-fire frames.
4. The 1DS-MIII has 14 bit A/D conversion.
5. The 5D-MII LCD has about 3 times more pixels than that of the 1DS-MIII.
6. The 1DS-MIII has about 3 times more AF points and almost twice as many light metering points.
7. The 1DS-MIII has more than twice as many customizable settings.
