If you are looking for luxury and high-rolling, I would suggest The Venetian or Bellagio over the Wynn.
You'll actually find that your choice of hotel to sleep in isn't that important. Except for use of the pool, you can spend your time in whichever casino you want.
The only places I've stayed that I would try to avoid staying in again are Circus Circus and Stratosphere, and that's mostly because of their location on the strip.

The best new hotel, with the best new technology. You room will greet you and light up upon entry. The curtains will open to the strip when you approach the windows, the television will turn itself on to familiarize you with the rooms' controls. If the internet, hybrid cars, and Project Natal aren't enough to convince you that you're living in the future, stay at Aria. It's Steve Wynn's newest casino. Vegas natives are familiar with his commitment to making everything larger-than-life and perfect, but visitors may not know it, thanks to his reduced focus on marketing the Wynn and Encore. Just know that this hotel is all about service, and exceeding expectations.
