Made in 2004, Anchorman stars Will Ferrell as Ron Burgundy, a 1970s news reporter who is hopelessly vain and idiotic. The supporting cast, which includes Christina Applegate, Steve Carell, Paul Rudd, and Fred Willard absolutely knocks this movie out of the ballpark.
But Anchorman is a SPECIAL movie. The first time you watch it, you won't like it. I promise. Everyone I know watched it once, gave a sour look and dismissed it. But on 2nd and 3rd viewings the movie becomes absolutely hysterical! Gno's husband raves: "Easily the funniest movie of the last 10 years!"
We've almost worn out our DVD. Never in my life have I quoted one movie SO much. And that's saying something!

M$In my opinion the funniest movie of this decade by far was the 2006 release Borat: Cultural Learnings of America for Make Benefit Glorious Nation of Kazakhstan

My first thought was Office Space, but that was made in 1999.
I think the last decade has highlighted a new brand of comedy called "Uncomfortable and Embarrassing". I think it started with movies like Meet the Parents (2000) and Meet the Fockers (2004) and branched out from there. The comedic actors that have ruled for the last decade are Ben Stiller, Will Ferrell and Vince Vaughn and their sidekicks.
Every decade brings a new generation and different skew to comedy, and I have found that you either like it or you don't. I have also found that you have to be in the right mood to take in a comedy. When I watched Napoleon Dynamite for the first time, I didn't like it, but watching it again... I enjoyed it more.
After scanning the decade list of comedy movies from the link above, I was able to narrow it down two movies, well... actually three that I found to be funny in a unique and quirky way. Two of these are not your typical cookie cutter comedies either.
Even though Snatch came out in 2000, I have to give it honorable mention.
I thought I Heart Huckabee's (2004) was a really different kind of comedy and if you didn't see it, I would highly recommend it.
And for plain and silly slapstick comedy, I would say Dodgeball: A True Underdog Story (2004) is also a good one.
