What is the best way to password protect a file on my desktop so that other people can't open it?

You could compress the file using a free program like 7zip and put a password on the archive. Here's an easy guide on how to do that.

As others have pointed out, you can use 7-Zip to password protect it, but if you're the paranoid type, you can also encrypt it with a free, simple utility called AxCrypt.

When encrypting, you are prompted for a password, and to view the file, you double-click it, enter the password and the file is opened.

If it's a Microsoft document, on the tools menu, there is an option to protect the document. It allows you to set a password.
