In a DoS attack, a computer (usually a web server) is flooded with so many connections that it cannot respond to legitimate requests.
Distributed Denial of Service (DDoS) attacks are similar, but rather than coming from a single location, requests are sent out from a variety of different locations. This makes the attack harder to stop.
Commonly, "botnets" are formed from millions of unsuspecting, virus infected personal computers and servers. Once activated, the combined power of the botnet is substantial and if properly activated can cause network administrators a little bit of trouble.
Sometimes, a website becomes so popular that without meaning to be an attack it floods a website with traffic causing it to stop working. This is commonly refereed to as the "Slashdot effect" or the "Digg effect".
