Posting a message does not, but refreshing your "stream" does and some applications (like Twitteriffic, Hahlo etc) that integrate DM's and @'s will hit the API 3 times for one refresh.
Currently you get 100 hits an hour, but when Twitter is being bombarded this can substantially drop to as low as 30

What actions affect the twitter API limit?
Clearly polling the API does, but I'm uncertain if posting a message also counts has a hit.
Does anyone know what actions affect which limits? Please provide me w/ limit name, limit total, and actions that affect the limit.

I remember hearing this somewhere, but for every external application (tweetdeck, twitterific, etc.) API REQUEST (so, every time the application refreshes your tweets) it uses one. According to the coutner on tweetdeck (my favorite client ;)) there are 100 requests per hour. So if you have your application set to refresh every minute, you will use 60 per hour. Now, if you leave that running, and you leave and use your iPhone's client to refresh, you will consume API requests as well, so if you refresh it once a minute on your iphone as well, you will hit the 100 request cap, and you won't be able to make any more requests for another hour. I don't think refreshing the twitter web page uses requests, so you can go crazy with that.
