The Ethereal Queen at the end of the Seraphic Gates is the most difficult boss, as she is the last boss that you fight - also, depending on the difficulty level, she is can be even more troublesome. Try beating the game on Infinity Mode, and get to the end of the Seraphic Gates - she'll be insanely difficult to defeat.
