What happens when you have beaten Lou on Medium? I saw something is unlocked, but I have no new songs and can see nothing unlocked!

It really depends on the level you finished what was unlocked. Here is what gets unlocked when you beat Lou ( finish a career) :

Moon Guitar Complete Easy mode.
Saint George Guitar Complete Medium mode.
Rojimbo! Guitar Complete Hard mode.
Risk Assessment Guitar Complete Expert mode.
Bat Guitar 5-star all songs in Easy mode.
Jolly Roger Guitar 5-star all songs in Medium mode.
Tiki Face Guitar 5-star all songs in Hard mode.
El Jefe Guitar 5-star all songs in Expert mode.
Pendulaxe Blade Bass Complete Co-op Expert Mode.
Assassin Bass 5-star Co-op Expert Mode.
Beach Life Bass Complete Co-Op Career Hard Mode.
Tiki Fortune 4 Bass Complete Co-op Easy mode.
The Disposed Bass 5 star Medium Co-op career

To play as Tom Morrello, Slash or Lou simply beat them then buy them.

The unlockable song is Through the Fire and Flames by DragonForce
To unlock you must beat the whole career mode on any difficulty.\

I think thats all of the unlockables not including the achievements.

Hmm according to this source you need to beat him in career mode and you get..

The source also mentions these tips for Easy and Expert mode, but nothing for medium unfortunately.

When the battle with Lou starts at the end of the game on Expert mode, strum without holding any frets until you lose. At the "Game Over" screen, select "Tutorial". Pick the tutorial for Guitar Battle, and play through it until the "Lesson Complete" message appears. As soon as the next fret board appears, pause the game, and exit to return to the lesson menu. Then, press Red to return to the list of songs. The battle with Lou will be highlighted. Choose the song, and Lou's notechart will now be set to Easy mode. Note: Your notechart will still be in Expert mode, but his will be in Easy mode.
