There is no "best hotel" in Las Vegas. If it's your first time, I would recommend MGM. It's huge, so you could probably spend your whole trip there and not get bored, but if you do, it's located next to a few other really good hotel/casinos.
If you are looking for luxury and high-rolling, I would suggest The Venetian or Bellagio over the Wynn.
You'll actually find that your choice of hotel to sleep in isn't that important. Except for use of the pool, you can spend your time in whichever casino you want.
The only places I've stayed that I would try to avoid staying in again are Circus Circus and Stratosphere, and that's mostly because of their location on the strip.

You didn't say if you were looking for luxary or good deal, but Trip Advisor lists Staybridge Suites Las Vegas as the "Best Value"
