i would say the greatest mystery know to man is The Shroud of Turin

I would consider this as the most mysterious of all here you have evidence and still today's technology cannot come up with answer.

I've always been intrigued by Jack the Ripper. It drives me nuts that they haven't solved this. I read a book by Patricia Cornwell a few years ago and she worked on resolving the case but I don't think they confirmed her suspicions. She just had a theory about who Jack was. I hope they continue to try to solve this case.
