I'm a big "Planes Trains and Automobiles" fan. I know it's sort of expected that everyone will say his classic high school comedies, but this is the one of his films that has always stuck with me, and I think it holds up really well. http://www.youtube.com/watch?v=NEZv0FUPtcc

This is one of my old time favorite movies period. I've done more business traveling then I care to remember. I laughed until I cried.

No point in just duplicating your answer because I completely agree! We watch it over and over every Thanksgiving season. Hilarious movie - even better than the Molly Ringwald flicks.
I'd also like to give a shout-out to Uncle Buck, a very underappreciated Hughes film with a lot of humor and a lot of heart.

So many great ones, but probably Weird Science, a film squarely aimed at geeky guys. Amazing how many other movies he wrote but didn't direct, like the Vacation, Home Alone and Beethoven films.
