Tell him you are interested in the job, add a cover letter to prove your
interest, and possibly even go as far as writing an essay on why you
would be so passionate about a job at Mahalo. Also, have references
prepared, and have them write why you should work at Mahalo.
Before you do any of this, I would first add some info to your own personal profile on Mahalo, as that shows you took the time to do so. For bonus points, put in your profile something like, "aspiring Mahalo employee" or something that's catchy enough to show you really have an interest in Mahalo.

The easiest way to do that is to ramp up your presence on Facebook, Twitter, Mahalo Answers. Then add Jason as a friend/follower/whatever. And add some other tech people with good networks.
Most of the other criteria seem to be things to show in an interview rather than beforehand, apart from showing that you can easily talk to all sorts of people. Perhaps get a video of you interviewing a startup CEO or someone similar, and then a homeless guy. And mash the videos together.

What is the best way to get an interview with Jason Calacanis?
Jason recently posted a job offering which I would love to have the chance to interview for. I feel I am a qualified candidate and I have sent him my cover letter and resume. I am curious if the community has any creative ideas on how to bring my resume to the top of his short list?

Send him a direct message on here... don't be afraid to talk with him! I would think the best way to get an interview would be to have a true passion for your work and to be able to communicate that in an open, pleasant and entertaining style. If you've got a good web presence (blog etc.), then make sure its relevant and informative and provide all of the information to Jason.

Assuming I'm talking about the same job offering, then I think the most important thing to demonstrate to Jason is your networking ability and to show you know something about technology.
