If you want to be as far south as you can in Florida, I'd recommend Marathon which is in the Florida Keys. http://www.catch-em-all.com/
It's not cheap to go deep sea fishing, but if you can find a large enough boat and go with a group, you'll be saving a pretty penny. Be prepared to pay $200 per person average.

M$Being the premier area for sportfishing, you can easily pay up to $1,000 per day per boat.
Of course this sounds like a lot of money, but if its what you really want to do then its best to get an expert who knows where the Marlin are in the water, its really this knowledge more than the hire of the boat that you are paying for,
Certainly around Islamorada and Key Largo you will find plenty of Captains ready to take you out.
