In your opinion would the US public be interested in a movie taking place in Africa if it would be more in coherence with the script?

Yes, of course. Many movies that take place in Africa have been popular with the US public!

If the story is good and the acting good then yes Americans would support this type of movie. Even if it was written for African's then it could find an audience here. Americans like movies made in and about other countries.

I'm not sure I understand what you mean by "more in coherence with the script". American movie goers for the most part pay money to see big stars in big action films. Of course, we all know that most of those movies are crap. If you have a script with powerful characters and an interesting story line, then I don't think the location is really an issue at all. I'm sure there are many films made in Africa.

The audience for independent films isn't as large as the audience that sees the mainstream movies. If your movie could get picked up and distributed by one of the bigger names then it might have a shot. However, I think independents only make it big if they win a major award and the film gets noticed. Sometimes those directors are then picked up by the studios.

So I think the answer to your question is yes, american audiences would see a film set in Africa but it would depend on the plot and the characters.
