He is unlocked by collecting all 120 stars available to Mario after you have defeated Bowser. Some stars are hidden until you defeat Bowser, they are signified by the Purple Comet.
I have unlocked Luigi myself, but I cannot speak towards the grand finale. Supposedly, there are 241 stars. 120 for Mario, the same 120 for Luigi, and then 1 last star to completely end the game. So basically you have to play all the way through twice. And you cannot use Mario to get Luigi's stars, you must collect all 120 with Luigi.
