They're all funny but Whoopi, Roseanne, and Tina Fey stand out because they've also contributed a lot to comedy writing, Fey as SNL head writer for the few brief years (counting recent times only) when that show was actually funny, plus on 30 Rock. For this reason I vote for Fey.
(There are of course a ton of other women - Wanda Sykes is hilarious, and Margaret Cho, Kathy Najimy, and Lily Tomlin have been influential, just to name a few).

Her performance in Ghost is one of the rare instances, when it got academy award as well as it made us laugh.
