A lot depends on your needs.
Are you asking from the viewpoint of a content creator/distributor? If you want sheer viewing numbers, YouTube has the largest audience, hands down. If you are looking for a more sophisticated audience, you may be better distributing your content on a "smaller" site such as Vimeo.
Is your target audience the "frat boy" demographic? Maybe Break.com or EJB, etc.
If you are asking, as a viewer of content, who has the best content, again, it depends on what you are interested in viewing. For sheer variety, YouTube can definitely satisfy. Want videogame movies? Gametrailers.com. Broadcast network shows like sitcoms and prime time? Hulu.com. Porn? Heh, well, you can't wave your arms on the internet without hitting a porn site, so take your pick.

If you want views, I'd recommend Youtube, simply because there are so many more people watching it, than any of the other video hosting sites. I can't actually even remember the last time I've even watched a video on Viddler or Vimeo. Also Youtube supports HD content now, so the old complaint of its video quality being low is now very much not true.
