Here are two in depth reviews of each phone from Gizmodo to help you determine for yourself.
iPhone 3GS : http://gizmodo.com/5293388/iphone-3gs-review
Palm Pre : http://gizmodo.com/5277499/palm-pre-review
I personally love my iPhone 3G and cant imagine switching to the Palm Pre. Plus I heard early Pre's had faulty construction and overheating issues.
On feel, I prefer the solid metal feel of the iPhone over the plastic flimsy feel of the Pre.
Also, as the iPhone is the best selling phone, it has the most people working to improve it in all ways, whether it be apps, jailbreak apps, etc.... I mean, the attention the iPhone gets is shown in the innovative ways to get around the stupid AT&T restrictions.

It might come down to one of these factors:
a) Which size and shape suits you? I'm happy with a bigger screen and a slimmer device, but your might prefer a smaller but chunkier device.
b) Over 50,000 apps are available for the iPhone. Chances of finding the one to do what you want are probably much better than on the Palm Pre at this point.
Both are cool devices, and I'd be happy to posses either.

Pre is better for seven reasons. 1)It feels small and pleasant in the hand
2)It runs linux
3)the animation and interface are gorgeous
4)got the coolest menu bar I have ever seen
5)interfacing with gmail and facebook look top notch
6) It has official application store and you can still download more apps

Some counter reasons:
1) The iPhone isn't that big and it also feels pleasant in the hand. Actually, the iPhone is slimmer than the Pre
2) Who cares about if it runs Linux? Most Palm devices run Windows Mobile and the iPhone runs Mac OSX mobile.
3)The iPhone also has an app store. Apple app store has over 50,000 apps while the pre has 12 and its store is still in a beta.
4)iPhone also has multitouch. In fact it was one of the first ones to have it.
5) Storage/price ratio: The pre comes in one size... 8GB and it will cost you $199 with a new contract AND a $100 mail-in-rebate. The iPhone comes in 8GB for the 3G and 16 or 32GB for the 3GS. The 8GB iPhone costs $99 with a new 2 year contract. The 16GB 3GS is $199 and the 32GB 3GS is $299 with new 2 year contracts.
6) Camera: Both have a 3 MP camera. The Pre has a flash but the iPhone 3GS has autofocus (select focus by touching area also) and the ability to record and edit video right on the phone. Then post it to YouTube.
7)iTunes. By far the iPhone has the advantage when it comes to iTunes. Yeah you can sync your Pre with iTunes but with the iPhone you can buy apps from the app store (which is part of iTunes), buy books, movies, tv episodes, music videos, songs, podcasts, and anything you can buy on your computer with iTunes you can do it with the iPhone and the next time you sync with iTunes it will transfer all those purchases to your computer.
Just some food for tought. I love my 32 GB iPhone 3GS

I don't know which is better for you, but there is a nice video on the iPhone 3G page that compares the two.
