Release when (if) the Apple logo appears on the screen.
If this doesn't work, and your iPhone is truly bricked, your only option is to return it to the shop you bought it from. The iPhone is capable of running 3.0(.1), so that shouldn't be the cause of the problem.

I have a 2G iphone that was running on 2.1 firmware. I accidently updated the software to 3.0.1 and now my iphone is bricked. Help!!

What do you mean by 'bricked'? Is it entirely dead, also after a hard reset? Try pressing and holding both the Home button and the Sleep/Wake button.
Release when (if) the Apple logo appears on the screen.
