This is a pretty strange question but I tried a few approaches to make an educated guess. Looking at the way humans knock each other out with a punch, that involves a punch to the head or face. Looking at the skull of a giraffe, there is no where that a punch could obviously be aimed for a good blow. Not the distant chin, not the sloping temple, not the horned (!) top of the head. Looking at the way giraffes fight, which is by swinging their long necks (head on the end remember) at each other, their heads must be pretty shock resistant. Finally, the average mass for an adult male giraffe is 1,191 kilograms (2,630 lb). That's big! Ten times the mass of a heavyweight, roughly.

I am assuming you are talking about a strong grown man familiar with boxing and a normal grown male giraffe. (no hitting girls and kids)

My conclusion is that it would be impossible. The giraffe is too massive and his head is well designed.
