If you're looking for a used car at the best price you can get, you should start with your local paper's classified ads. Usually the sellers there are looking to get rid of their used cars as soon as possible and will probably be willing to lower their initial asking price.
Will you get the best used car for your money this way? Maybe, but it's best if you buy yourself a little insurance anyway. Ask the seller if they would mind you taking their car to an independent mechanic. Pay the mechanic $50 or so to check it out thoroughly before agreeing to any price. You'll be glad you did.
But if you want a guarantee of getting a good used car, check out something like Carmax. They go over every car they sell before putting it on their lot. Sure, the car will end up costing more because of that process, but you can sleep a little easier knowing it's been thoroughly inspected.
If you're talking about new cars, you can use Edmunds.com or Consumer Reports to determine how much on average people are paying for the model you're interested in.
