The first woman to actually work on the floor was Helen Hanzelin.
In the links below I have included the link to Muriel's company (Muriel Siebert & Co.), the Wikipedia reference, the Britannica reference and extended interview outtakes from her NBC Nightly New interview.
There is less on Helen Hanzelin. The links below include yahoo answers, and a reference from Google book, A Financial History of the United States.

Who was the first woman to set foot on Wall street trading floor in 1943?
I've been told that my cousin, Athlene Grayson (possibly Rogers at that time) was the first women to set foot on the floor of Wall Street?

The first woman to hold a seat on the NYSE is Muriel Siebert. The first woman to actually work on the floor was Helen Hanzelin.
