If it has been published and you just want to get it read, then your best bet is approaching local bookstores to help do a feature display or a 'meet the writer' (don't charge them and they will be more willing to help you out! But off a 'finder's fee' % to the store of all sales) You can submit to critics of papers or magazines, and hope they give you a helping hand with a good review. You will have to sacrifice a bit of sales to make sure some copies get into the hands of people you feel could help you. Think of where YOU would find info about good books, and make sure your stuff is put out there too. Also hiring a PR/marketing (if $$ isn't a factor) can help you out as well (it's what they do best). But today with Facebook, Myspace, Twitter etc, word of mouth would do anything some good. make sure the right people read it, and then they tell their friends and they tell their friends...
If is hasn't been published, then you have to submit to publishing houses directly. The bigger the firm the harder it will be to get published, but they will also be able to bring you marketing and publicity support (they want to make their money too, but be prepared for editors from hell) because of their reputation. Smaller firms will be more likely to publish, but my not have the best publicity (but you can also deal with that through another person/agency not associated with the publisher) Submit to both anyway, see who nibbles and make your decisions from there. But remember the more doors you knock on the more likely you are to have it answered (it's the law of average). And don't stop until it's published.

The traditional route is to try to get reviewers to cover it and get on talk shows to promote - you will probably have to have a publisher to make that route happen.
Another route is self-promotion. Get it listed on Amazon and try to get bloggers to talk about your book. Get your book listed on your web site (start one if you don't have one).
Good luck.

There is no "best" way per se, because it depends upon the genre of the book, but you can talk to the owner of this publishing outfit about it if you're curious, because she's been through it all. http://driftwillowpress.com

The topic of your book may play a role in your marketing strategy. The traditional route is to try to get reviewers to cover it and get on talk shows to promote - you will probably have to have a publisher to make that route happen.
